An antibody specific for GHRH has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyGHRH present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for GHRH is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GHRH bound in the initial step. GHRH is released from neurosecretory nerve terminals of these arcuate neurons, and is carried by the hypothalamo-hypophyseal portal system to the anterior pituitary gland where it stimµLates growth hormone (GH) secretion by stimµLating the growth hormone releasing hormone receptor.
